N2 - Anticonvulsant effect of 8α,9α- and 8β,9β-epoxyhexahydrocannabinols (EHHCs) on pentylenetetrazol (PTZ)-induced seizures was examined in mice and compared with that of Δ8-tetrahydrocannabinol (THC). After injection of PTZ, latency of clonic and tonic convulsions was longer in mice pretreated with these cannabinoids than that in the vehicle-pretreated control group. 8β,9β-EHHC was most effective in increasing latency, followed by Δ8-THC and 8α,9α-EHHC. Furthermore, most of the mice pretreated with cannabinoids did not show lethal tonic extension. In combination with phenobarbital, percent protection of tonic convulsion tended to be higher in 8β,9β-EHHC-pretreated mice than that in the group pretreated with other cannabinoids. From these results, the potency of anticonvulsant activity of the cannabinoids tested against PTZ-induced seizures was ranked in the following order; 8β,9β-EHHC > Δ8-THC > 8α,9α-EHHC.
