Beer and prevention of heterocyclic amine-induced DNA adducts and O6-methylguanine

Non-volatile dissolved materials in beer amount to as much as about 5% (w/w) on freeze-drying of beer samples. These dissolved components should be taken into account for any study of the biological effects of beer. We have described here inhibitory effects of beer on the formation of O6-methylguanine (O6-meG) by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the DNA of Salmonella YG7108 when beer was added to bacteria in the presence of MNNG. We have also evaluated the in vivo effects of beer components on the formation of DNA adducts by Trp-P-2, MeIQx and PhIP under conditions reflecting human dietary habits. The protective effects were found in liver and lung, the target organ for MeIQx tumorigenesis in mice, with a drinking solution containing beer components and also by adding beer components to the diet to simulate intake of food additives. We also investigated the inhibitory effects of a solution of beer components on DNA damage in mice liver with single and continuous administration of Trp-P-2. Furthermore, we evaluated the in vivo protective effect of beer samples in relation to PhIP-induced DNA adduct formation in the colon (the target organ associated with PhIP tumorigenesis) and other important organs (liver, lung and kidney) of mice fed with a beer solution or by the addition of beer components to the diet.
