Heat stability of Y-chromatin in blood smears

Abstract

The influence of high temperature on Y chromosome with respect to quinacrine stainings was studied. Blood smears from 5 males and 3 females were tested in this study. Results showed that in male blood Y-chromatins could be detected in more than 10% of nuclei in the leucocytes by quinacrine staining after heating at 100°C for 5h, at 200°C for 60 min and at 300°C for 60s, while in female blood under the same condition Y-chromatins could be detected in 1-2% of nuclei and therefore sex identification could be achieved. This study indicates that Y-chromatin is heat stable.
