Identification and determination of 8α9αepoxyhexahydrocannabinol as a metabolite of δ8-tetrahydrocannabinol of mice in vitro

Abstract

1. 8α9αEpoxyhexahydrocannabinol (8α9αEHHC) was formed from δ8-tetrahydro-cannabinol (δ8-THC) by mouse liver microsomal preparation in the presence of an Nadph-generating system. 2. The epoxide was identified by t.l.c., g.l.c. and g.l.c.-mass spectrometry and, together with 11-hydroxy-δ8-THC (11-OH-δ8-THC), was determined by g.l.c. 3. When δ8-THC was incubated with mouse liver microsomal preparation, 8α9αEHHC and 11-OH-δ8-THC was formed to the extents of 14% and 23% respectively, of the added δ8-THC.
