Transdifferentiation of retinal pigment epithelial cells into lens in suspension culture

Purpose. To elucidate factors influencing transdifferentiation of retinal pigment epithelial (RPE) cells in suspension culture. Methods. RPE eel Is were isolated as a sheet from 7.5-day-old chick embryos and cultured either as a sheet or dissociated eelIs after trypsin treatment in a non-adherent dish for suspension culture for one month. Basic fibroblast growth factor (bFGF) was added to the medium at varying concentrations of 0-200 ng/ml. Results. At the higher concentrations (10 or 200 ng/ml) of bFGF, a rounded-up RPE sheet grew to develop a nonpigmented lump with an onion-like structure, while dissociated RPE cells, although they reaggregated, did not form such structure. Histologically, the onion-like structure consisted of lens-fiber-like cells, positive for ant i-crystall in antibody and anti-bFGF-receptor antibody by immunohistochemistry. The formation of this lens-1ike structure did not occur at the concentration of bFGF, 1 ng/ml or lower. Conclusion. The transdifferentiation of RPE cells into lens in suspension culture was dependent upon the concentration of bFGF as well as proper three-dimensional interaction among each RPE cell.
